ارزیابی میزان تغییرات بیان ژن های خانه دار در برهمکنش گندم با بیمارگر Mycosphaerella graminicola با روش Reverse northern dot blot
بهطور کلی ژنهای مرجع در تجزیه و تحلیلهای بیان ژن، از بین ژنهای خانهدار انتخاب میشوند، اما تغییرات دیده شده در مقدار بیان مانع اساسی برای استفاده بهینه از آنهاست. از آنجا که بیان هیچ تک ژنی در شرایط مختلف و سلولهای متفاوت ثابت نیست، معرفی و اعتبارسنجی ژنهای مرجع بسیار مهم است. در این تحقیق مناسب بودن هفت ژن خانهدار گندم برای نرمالسازی بیان mRNA در برگ این گیاه در هنگام آلودگی به قارچ Mycosphaerella graminicola مطالعه شده است. به این منظور بیان ژنهای رمزکنندۀ اکتین، روبیسکو، گلیسرآلدهید 3- فسفات دهیدروژناز ، فاکتور طویلسازی ترجمۀ 1 آلفا (TEF 1α)، آلفا-توبولین، فاکتورهای آزادکنندۀ یوکاریوتی 1 و 3 (ERF1 و ERF3) در طول آلودگی توسط روش نوردن بلات معکوس اندازه گیری، و توسط نرم افزارهای اکسل و SAS از لحاظ آماری بررسی شد. بیان ژنهای آلفا-توبولین، TEF 1α و اکتین از بیشترین پایداری برخوردار بود و ژنهای روبیسکو و گلیسرآلدهید 3- فسفات دهیدروژناز کمترین ثبات بیان را داشتند. مطالعه مقایسهای تغییرات در بیان ژنهای مختلف خانهدار گندم در پاتوسیستم گندم - M. graminicola انتخاب ژنهای مرجع را برای روش لکهگذاری نوردنبلات معکوس تسهیل میکند.

بیان ژنهای خانواده CYP دخیل در سنتز سزامین در دانه ارقام مختلف کنجد (Sesamum indicum L)
کنجد (L. Sesamum indicum) با داشتن مقادیر زیاد آنتیاکسیدان و متابولیت ثانویه منبع تغذیهای مهمی در تامین و حفظ سلامت انسان است. اهمیت ویژه دانه کنجد بهدلیل وجود آنتیاکسیدان طبیعی مهمی به نام سزامین است. بهدلیل کاربرد این آنتیاکسیدانت در پیشگیری از بروز بیماریها بهخصوص سرطانها تلاشهای زیادی در راستای شناخت ییشتر ژنهای دخیل در مسیر بیوسنتز این ماده و افزایش سطح بیان ژنهای رمز کننده آنزیمهای مربوطه انجام شده است. در این مطالعه سطح بیان ژنهای کلیدیCYP81Q1 ،CYP81Q2 ،CYP81Q3 و C3H دخیل در بیوسنتز سزامین در دانه 10 رقم کنجد شامل رقم یکتا، زودرس فلسطینی، ناز تک شاخه، ناز چند شاخه، اولتان، جیرفت، دشتستان 5، دشتستان 2، ورامین و کرج 1 با روش QRT-PCR و ژن خانهدار 18S rRNA بررسی شد. براساس نتایج حاصله سطح بیان ژن CYP81Q1 در رقم زودرس فلسطینی پایین بوده و بعلت پایین بودن مقدار سزامین آن، این رقم را نمیتوان در مقایسه با سایر ارقام بهعنوان یک رقم روغنی مطلوب از نظر کیفییت معرفی کرد. رقم یکتا در بین ارقام بیشترین سطح بیان ژن CYP81Q1 را داشت و یکی از بهترین ارقام در تولید روغن از نظر کیفی و کمی است. ژن CYP81Q2 در رقم ناز چند شاخه بیشترین و ژن CYP81Q3 در رقم زودرس فلسطینی کمترین سطح بیان را داشتند. همچنین ارقام کرج 1، یکتا و دشتستان 5 بیشترین سطح بیان ژن C3H و اولتان کمترین سطح بیان را داشتند. ارقام کرج 1، یکتا و دشتستان 5 جزء ارقام تقریباً دیررس هستند. ارقام دیررس دارای روغن بیشتر و محتوای سزامین بالاتری در مقایسه با ارقام زودرس هستند.

بررسی بیوانفورماتیکی و بیان خانواده ژنی SnRK2 در گیاه جو
تنشهای غیرزیستی از مهمترین عوامل محدود کنندهی عملکرد گیاهان زراعی و پروتئین کینازهای SnRK2 از تنظیمکنندههای کلیدی پاسخ گیاهان به تنشهای غیرزیستی میباشند. با توجه به اهمیت اقتصادی، سطح زیر کشت و تحمل گیاه جو به تنشهای غیرزیستی، در این تحقیق اعضای خانوادهی SnRK2 در گیاه جو شناسایی و مورد بررسی قرار گرفتهاند. بههمین منظور توالیهای حفظ شده خانواده SnRK2 در پایگاههای اطلاعاتی مختلف همچون NCBI و پروژه ژنوم جو، با ابزار tBLASTn در بین توالیهای ثبت شده برای گیاه جو جستجو شدند. در نتیجه 10 عضو یافت شدند HvSnRK2.1) تا HvSnRK2.10) که 8 عضو آن تاکنون گزارش نشده بودند. اعضای خانوادهی SnRK2 جو با اعضای این خانواده در آرابیدوپسیس و برنج همردیف و درخت فیلوژنتیک رسم شد. تعیین جایگاه کروموزومی، آنالیز پروموتر و تعریف ساختار ژنها انجام شد و الگوی بیان هر ژن در اندامها، تیمارها و ارقام مختلف بر اساس دادههای ریزآرایه مورد بررسی قرار گرفت. نیمی از اعضای این خانواده روی کروموزوم 2 و بقیه روی کروموزومهای 1، 4، 5 و 6 قرار داشتند. تعداد اینترون در اعضای این خانواده از صفر تا 8 متغیر بود. 19 نوع عامل سیس شامل عوامل مؤثر در پاسخ به تنشهای غیرزیستی، بر روی پروموتر اعضای خانواده ژنی HvSnRK2 شناخته شد. بیان ژن HvSnRK2.6 توسط خشکی، شوری و سرما القا شده و بهنظر میرسد در انتقال پیام این تنشها و ایجاد تحمل به آنها نقش مهمی ایفا کند. امید است بتوان از این ژن برای اصلاح و دستورزی گیاهان در جهت تحمل به تنشهای غیرزیستی بهویژه خشکی بهره برد.

بهمنظور بررسی تنوع ژنتیکی 68 ژنوتیپ آفتابگردان از نشانگر جدید مولکولی TRAP با 6 آغازگر ثابت و 6 آغازگر تصادفی و 19 ترکیب آغازگری استفاده شد. تمام ترکیبهای آغازگری چند شکل بودند که در مجموع 116 باند ایجاد کردند که 109 تای آنها چندشکل بود. در این تحقیق لاینهای برگرداننده باروری با میانگین 76/22 باند چندشکل، بیشترین جایگاه چند شکل (48/84 درصد) و هیبریدهای ایرانی با میانگین 97/2 باند چندشکل، کمترین جایگاه چند شکل (52/40 درصد) را بهخود اختصاص دادند. بیشترین فاصله ژنتیکی بین گروه برگرداننده باروری و هیبریدهای ایرانی (151/0) و کمترین فاصله بین هیبریدهای خارجی و ارقام آزاد گردهافشان خارجی (064/0) بود. گروه لاینهای برگرداننده باروری و مادری کمترین فاصله ژنتیکی را داشتند (066/0). بر اساس تجزیه واریانس مولکولی، 87 درصد از تنوع ژنتیکی ناشی از تنوع درون گروهها و 13 درصد مربوط به تنوع بین گروهها بود. در هر دو تجزیه خوشهای و هماهنگکنندههای اصلی، گروه لاینهای برگرداننده باروری و مادری در یک گروه، هیبریدها و ارقام خارجی در گروه مشابه و هیبریدهای ایرانی در گروه مجزا قرار گرفتند. بر اساس تجزیه خوشهای کلی، پایینترین ضریب تشابه (472/0) بین لاینهای R42 و CMS328 محاسبه شد. نتایج نشان داد که نشانگر TRAP کارایی بسیار خوبی در گروهبندی و محاسبه تنوع ژنتیکی آفتابگردان دارد. با وجود تنوع ژنتیکی میان گروهها و ژنوتیپها، بهدلیل ضریب تشابه ژنتیکی بالای ژنوتیپها (755/0)، تنوع ژنتیکی ژنوتیپهای مورد بررسی پایین برآورد شد و بدینوسیله افزایش تنوع ژنتیکی ژرپلاسم آفتابگردان و انتخاب لاینهای اینبرد والدینی جدید با تنوع بالا در برنامههای آینده اصلاح آفتابگردان پیشنهاد میشود.

استخراج مستقیم و خالص سازی DNA کل از جامعه میکروبی خاک و ردیابی مولکولی فرمهای اختصاصی بیمارگر خاکزاد Fusarium oxysporum عامل پژمردگی آوندی گوجه فرنگی
دستیابی به اطلاعات مولکولی و ژنتیکی توانسته است بسیاری از چالشهای علوم زیستی را در جنبههای مختلف حل کند. روشهای کاربردی بسیاری جهت استخراج DNA از بافتهای جانوری و گیاهی و سایر موجودات زنده شناسایی و معرفی شده است.، در بسیاری مطالعات ژنتیکی در جوامع میکروبی، اغلب نیاز به استخراج مستقیم DNA از خاک است. در این تحقیق پس از مطالعه دقیق روشهای مختلف پیشنهادی، سه روش (شیمیایی- آنزیمی)، (شیمیایی- مکانیکی) و (شیمیایی- آنزیمی- مکانیکی)، جهت استخراج DNA ازجامعه میکروبی خاک، جداگانه طرح و مورد مطالعه و بررسی قرار گرفت تا مناسبترین روش، شناسایی و معرفی شود. مقایسه بین روشهای مختلف در شرایط برابر و 5 تکرار برای هر روش، در محصول DNA اختلاف مشخصی را از میانگین غلظت32/2 تا 1/6 نانوگرم بر میکرولیتر DNA رقیق شده، نشان داد و همچنین بر اساس اطلاعات بدست آمده از دستگاه نانودراپ و الکتروفورز و بررسی نسبت DNA به پروتئین در محصول استخراج DNA، روش (شیمیایی- آنزیمی-مکانیکی) با کارایی بهتر در غلظت بیشتر، حذف هیومیکاسید، پروتئین و سایر آلودگیها به سایر روشها برتری داشت. در این تحقیق همچنین ردیابی مولکولی بیمارگرها به عنوان یکی از کاربردهای مهم استخراج مطرح و فرم اختصاصی Fusarium oxysporum f. sp lycopersici و Fusarium oxysporum f. sp. radicis lycopersici عامل پژمردگی آوندی در گوجهفرنگی از جامعه میکروبی خاک به وسیله تکنیکهای مولکولی با آغازگر اختصاصی uni شناسایی شد. نتایج ردیابی مولکولی، حضور فرمهای اختصاصی این بیمارگر را در نمونه خاک S1 با 5 تکرار تائید مینماید.

فرابیان ژن شبه استریکتوسیدین سینتاز-6 در گیاه Arabidopsis thliana
استریکتوسیدین سینتاز یک آنزیم کلیدی مسییر بیوسنتز ایندول الکالوئیدهای مونوترپنی میباشد. آرابیدوپسیس قادر به تولید ترکیبات آلکالوئیدی نیست اما، ژنهایی مشابه استریکتوسیدین سینتاز در آن شناسایی شده است. ژن SSL6 از اعضاء خانواده ژنی شبه استریکتوسیدین سینتاز آرابیدوپسیس میباشد که در این تحقیق پاسخ ژن آن به تیمار شوری بصورت کمی بررسی و سپس در گیاه آرابیدوپسیس فرابیان شد. بمنظور تولید موتانت فرابیان ژن SSL6 RNA کل استخراج و رشته اول cDNA با استفاده از آغازگرهای اختصاصی ژن ساخته شد. کلون سازی ابتدایی در ناقل pJET انجام و متعاقبا به سازه فرابیانی گیاهی (pPZPY:SSL6) منتقل شد. در ترانسفورماسیون آرابیدوپسیس از اگروباکتریوم نژاد GV3101(PMP90) و تکنیک غوطهوری گلآذین استفاده شد. آنالیز گیاهان تراریخت احتمالی در ابتدا با کشت بذرهای حاصل از گیاهان تلقیح شده بر روی محیط گزینشی حاوی آنتی بیوتیک جنتامایسین و گزینش گیاهچههای مقاوم انجام شد. در ادامه گیاهان تراریخت احتمالی در طی واکنش PCR با پرایمرهای اختصاصی ژن SSL6 با تولید الگوهای باندی متفاوت در الکتروفورز در مقایسه با گیاهان وحشی و همچنین پرایمرهای اختصاصی ژن مارکر جنتامایسین جداسازی شدند. میزان افزایش بیان ژن انتقال یافته نسبت به گیاه مادری با واکنش Real-Time PCR بر روی گیاهان نسل T2 انجام و نتایج نشان داد افزایش بیان ژن SSL6 در گیاهان تراریخته به طور قابل ملاحظهای بالاتر از گیاه مادری بوده است. تعدادی لاین تراریخت حاوی یک نسخه تراژن از طریق تفرق صفت مقاومت به آنتیبیوتیک جنتامایسین در محیط کشت انتخابی در نسل T2 و T3 انتخاب شدند. بررسی کمی بیان ژن SSL6 در گیاهان وحشی Col-0 نیز نشان داد که تنش شوری موجب افزایش معنیدار بیان ژن، شش ساعت پس از اعمال تیمار شد.
