پاسخ های فیزیولوژیکی و متابولیکی گیاه برنج تحت شرایط تنش شوری
دو ژنوتیپ برنج IR29 و FL478با حساسیت های متفاوت به شوری تحت تنش شوری با 100 میلی مولار NaCl برای پاسخ های فیزیولوژیکی و متابولیکی مورد بررسی قرار گرفتند. به این ترتیب وزن خشک، وزن تر و طول ریشه و اندام هوایی گیاهان و همچنین میزان یون های سدیم و پتاسیم، مقدار اسید های آمینه و برخی قند ها و قند- الکل ها در ریشه و اندام هوایی گیاهان در شرایط کنترل و تنش ارزیابی شدند. بعد از 12 روز از تنش، سطح یون سدیم بخصوص در اندام هوایی IR29 افزایش معنی داری داشت در حالیکه یون پتاسیم بیشترین افزایش را در ژنوتیپ FL478 نشان داد. از طرف دیگر تغییرات متابولیکی در پاسخ به تنش های شوری در بین دو ژنوتیپ متفاوت بود، یعنی گیاهان IR29 در مقایسه با FL478 بیشترین سطح تغییرات را در اسید های آمینه بخصوص آسپارژین، گلوتامین، پرولین و GABA نشان دادند، این امر می تواند نشانگر خسارت و پیری سلول ها در گیاه حساس باشند، در حالی که قند ها و قند- الکل ها که به عنوان مواد محافظتی در سلول، تحت شرایط تنش در هر دو ژنوتیپ افزایش یافت، اما مقدار آن ها در ژنوتیپ متحمل FL478 بیشتر بود. این نتایج نشان می دهد که تفاوت های مشاهده شده در بین دو ژنوتیپ در پاسخ به تنش شوری ممکن است تا حدوی به تفاوت های مشاهده شده در سطح متابولیت ها نسبت داده شود.

بررسی تنوع ژنتیکی ارقام برنج ایرانی با استفاده از نشانگرهای ریزماهواره پیوسته با ژن های مقاومت به خشکی
چکیدهاین تحقیق به منظور بررسی تنوع ژنتیکی مجموعه ای از 20 رقم برنج ایرانی، با استفاده از 19 نشانگر ریز ماهواره پیوسته با ژنهای کنترلکننده تحمل به خشکی، انجام گرفت. همچنین شاخص مقاومت به خشکی به عنوان یک صفت تکمیلی مهم در مرحله رسیدگی دانه در 20 رقم و دو محیط (تحت تنش و بدون تنش) ، مورد ارزیابی قرار گرفت. نتایج تجزیه واریانس مرکب نشان داد که بین ژنوتیپها از لحاظ صفت شاخص مقاومت به خشکی، اختلاف معنیداری (در سطح احتمال یک درصد) وجود داشته و اثر محیط نیز بر شاخص مقاومت به خشکی در سطح احتمال یک درصد معنیدار بود. بر اساس نتایج آزمایش حاضر رقمهای سنگجو ، خزر و لاین 831 بیشترین پایداری عملکرد و ویژگیهای فیزیولوژیکی را در برابر تنش کمبود آب نشان دادند. همه 19 نشانگر ریز ماهواره در 20 ژنوتیپ مورد مطالعه چند شکلی نشان دادند. در مجموع 142 آلل با میانگین 47/7 آلل در هر جایگاه ژنی مشاهده شد. نشانگر RM166 دارای بیشترین تعداد آلل(11آلل) و نشانگرهای RM152 و RM555 دارای کمترین تعداد آلل (5آلل) بودند. همچنین میانگین PIC نیز 817/0 برآورد شد که نشانگر RM166 با PIC 89/0 بیبشترین و RM152 با PIC 7/0 کمترین PIC را نشان داد. محاسبه شباهت ژنتیکی بین دادههای مولکولی توسط ضریب تشابه Jacardو الگوریتم UPGMA، ژنوتیپها را به 6دسته تقسیم کرد. تجزیه به مختصات اصلی به نحوی تأیید کننده گروه بندی تجزیه خوشه ای ژنوتیپ ها بود. تمامی نشانگر های ریزماهواره مورد استفاده در این تحقیق تنوع ژنتیکی بین ارقام برنج ایرانی را نشان دادند.

بررسی بیان ژن های AlSOS1 و AlNHX در گیاه هالوفیت Aeluropus littoralis Parl. تحت تنش کلرید سدیم
شوری و به طور عمده کلرید سدیم، از رشد گیاهان جلوگیری نموده و سبب کاهش تولیدات کشاورزی میگردد. در گیاهان عالی دفع و حجره بندی سدیم بواسطه آنتی پورترهای موجود در غشاهای پلاسمایی و واکوئلی انجام می شود. در این مطالعه، الگوی بیان ژنهای AlNHX و AlSOS1 در پاسخ به تیمار شوری 250 میلی مولار کلرید سدیم در زمان های 6 ساعت، 1، 3، 8 و 17 روز پس از اعمال تنش با استفاده از تکنیک Real Time-PCR در گیاه Auleropus مورد بررسی قرار گرفت. سطوح نسخه برداری دو ژن در پاسخ به تنش در همه بافت ها افزایش یافت. بیان ژن AlSOS1 در بافت برگ پس از 6 ساعت افزایش یافت و بیان ژنAlNHX پس از 24 ساعت از اعمال تنش به بالاترین میزان خود رسید. در بافت گره و میانگره سطوح نسخه برداری هر دو ژن، 24 ساعت پس از اعمال تنش به شدت افزایش یافت و سپس در 3 و 8 روز بعد از اعمال تنش به تدریج کاهش یافت تا در نهایت 17 روز پس از تنش به حالت پایدار برابر با شاهد(بدون تنش)بازگشت. میزان بیان هر دو ژن در بافت های ریشه به آهستگی بعد از اعمال تنش افزایش یافت و پس از 3 روز به میزان حداکثر رسید و این میزان بیان تا 8 روز پس از تنش ادامه یافت و در ژن AlNHX پس از 17 روز به حالت پایدار برابر با شاهد بازگشت، در حالی که در مورد ژن AlSOS1 پس از 17 روز همچنان بیان دو برابر شاهد بود

انتقال ژن EXPA1 به گیاه آرابیدوپسیس تالیانا از طریق غوطه وری گل آذین
خشکی مهمترین تنش محیطی است که تاکنون به محصولات کشاورزی خسارت وارد کرده است. تاکنون تلاشهای زیادی در جهت بهبود محصول در شرایط کمبود آب صورت گرفته است.اکسپنسین ها یک ابرخانواده پروتئینی هستند که در گیاهان آوندی از چهار خانواده تشکیل شدهاند. اکسپنسین ها دستهای از پروتئینهای دیواره سلولی هستند که زمینه نرم شدن وابسته به اسیدیته دیواره سلولی از طریق شکستن پیوندهای هیدروژنی بین سلولز و شبکه گلیکان ها را فراهم می کنند. در این تحقیق برای بدست آوردن گیاهان تراریختی که دارای ریشه توسعه یافته تری باشند ابتدااز گیاهآرابیدوپسیس RNA استخراج گردید و پس از ساخت cDNA رشته اول و سپس تکثیر آن از طریق اغازگرهای اختصاصی برای ژن EXPA1در مرحله همسانه سازی، سازه ژنی pBIEXPA1 ساخته شد. از این سازه که حاوی ژن nptII و ژن AtEXPA1 تحت کنترل راه انداز CaMV35s بود در مرحله انتقال ژن جهت تراریختی آرابیدوپسیس استفاده گردید. عمل تراریختی با استفاده از آگروباکتریوم و تکنیک غوطه وری گل آذین صورت گرفت که این تکنیک به مراحل وقت گیر کشت بافت نیاز ندارد و گیاهان بدست آمده از آن نسل T1میباشند. در نتیجه وارد شدن سازه ژنی به برخی از بذرها، گیاهچههای حاصل از آنها بر روی محیط حاوی 50 میلی گرم در لیتر کانامایسین سبز باقی ماندند. تراریختی گیاهان بدست آمده با استفاده از واکنش زنجیرهای پلیمراز در سطح DNA تائید گردید. طبق انتظار دو باند 753 و 1080 جفت بازی در گیاهان تراریخت مشاهده گردید

ترانسفورماسیون پایدار و آسان جنین بالغ برنج (Oryza sativa L. var. indica) با استفاده از روش In planta
ترانسفورماسیون ژنتیکی برنج با استفاده از اگروباکتریوم (Agrobacterium tumefaciens) رویکردی مطلوب، ضروری و قدرتمند برای انتقال ژن به گیاهان زراعی میباشد. در این پژوهش به منظور ترانسفورماسیون گیاهان برنج از روش in planta استفاده شد. بر این اساس، بذور برنج به مدت دو روز خیسانده شدند و سپس سمت مریستم انتهایی بذر که جنین بالغ برنج وجود دارد در مراحل اولیه جوانهزنی با سوزن آغشته به محلول اگروباکتریوم زخم زنی و تلقیح -شد. آزمایش با دو سویهی اگروباکتریوم ( EHA105و LBA4404) حاوی پلاسمید pCAMBIAl105.1R، سه غلظت استوسیرینگون (Mμ200، 100 و 0)، سه رقم برنج (هاشمی، حسنی و غریب) و دو روش وکیوم و بدون وکیوم به صورت فاکتوریل و در قالب طرح کاملاً تصادفی در سه تکرار انجام شد. کارایی ترانسفورماسیون با استفاده از آزمون مقاومت بافتهای برگی به آنتیبیوتیک هایگرومایسین، آزمون بافت شیمیایی GUS و واکنش PCR با حداقل سه ژن مختلف بررسی شد. در نهایت، سویهی EHA105 در رقم هاشمی، با حضور Mμ100 استوسرینگون همراه با استفاده از وکیوم بالاترین کارایی ترانسفورماسیون (46/37%) را نشان دادند. پایداری ترانسژن در نسل بعد بررسی و بدین منظور 60 گیاه از نسل T1نیز با استفاده از سه تست اشاره شده آزمون و ترانسفورماسیون بودن 21% آنها تأیید شد.

بررسی بیوانفورماتیکی نقش جایگاه آمینو اسیدی 134 در اختصاصیت سوبسترای آنزیم دی هیدروفلاونول 4-ریدوکتاز دخیل در بیوسنتز آنتوسیانین ها
رنگ گیاهان اساسا ناشی از سه دسته ترکیبات فلاونوئید ها، کاروتنوئید ها و بتالین ها می باشد. فلاونوئید ها و بطور خاص آنتوسیانین ها رنگریزه های اصلی گل ها، میوه ها و بذر هستند. سه آنتوسیانین سیانیدین، پلارگونیدین و دلفینیدین آنتوسیانین های اصلی در گیاهان می باشند. مسیر بیوسنتزی منتهی به آنتوسیانین ها در بین گونه های مختلف بخوبی محافظت شده است. آنزیم دی هیدروفلاونول 4-ریدوکتاز یکی از آنزیم های کلیدی در مسیر بیوسنتزی آنتوسیانین ها است که دی هیدرو فلاونول ها را به لئوکوآنتوسیانیدین های مربوطه تبدیل می کند. برای بررسی نقش جایگاه آمینو اسیدی 134 آنزیم دی هیدروفلاونول 4-ریدوکتاز در تعیین اختصاصیت سوبسترای آنزیم در گونه های مختلف، توالی های آمینو اسیدی این آنزیم را از پایگاه های اطلاعاتی جمع آوری کرده و مورد تجزیه و تحلیل قرار داده ایم. نتایج هم ردیفی چندگانه آمینو اسیدی نشان داد که توالی آمینو اسیدی آنزیم دی هیدروفلاونول 4-ریدوکتاز در بین گونه های مختلف محافظت شده می باشد. در دو گونه از گونه های مورد بررسی هیچکدام از اسید آمینه های محافظت شده آسپارژین و آسپارتیک اسید در این جایگاه وجود نداشته است. نتایج ما به وضوح نشان داده که این جایگاه به تنهایی نمی تواند مسئول تعیین اختصاصیت آنزیم دی هیدروفلاونول 4-ریدوکتاز باشد و جایگاه های آمینو اسیدی مجاور جایگاه 134نیز ممکن است در این تعیین اختصاصیت سهیم باشند.

بررسی ایمنولوژیک و عملکرد استیل ترانسفراز مخمری (AYT1) در سم زدایی مایکوتوکسین دی اکسی نیوالنول در گیاهان توتون تراریخت
بیماری بلایت فوزاریومی سنبله گندم که به وسیله قارچ Fusarium graminearum ایجاد می شود, قادر است علاوه بر کاهش چشمگیر عملکرد, خسارت غیر مستقیمی را نیز از طریق تجمع مایکوتوکسین ها (تریکوتسین ها) در دانه های برداشت شده وارد سازد که محصول را برای تغذیه انسان و دام نامناسب می نماید. تریکوتسین ها (مانند دی اکسی نیوالنول یا DON) ممانعت کنندگان سنتز پروتئین در یوکاریوت ها می باشند. محصول ژن AYT1 در مخمر S. cerevisiaeتوانایی 3- O- استیلاسیون تریکوتسین ها را داراست و می تواند DON را به فرم استیله که از سمیت پایین تری برخوردار است تبدیل کند. استیلاسیون یکی از مکانیسم های سم زدایی و ایجاد مقاومت به مایکوتوکسین هاست, در این مطالعه AYT1 (استیل ترانس فراز مخمری) با استفاده از اگروباکتریوم به گیاه مدل توتون انتقال داده شد تا امکان ایجاد مقاومت نسبی به بیماریFHB, مورد بررسی قرار گیرد. به منظور سهولت پیگیری بیان تراژن مذکور اپی توپ c-Myc با استفاده از تکنیک PCR-Tagging به آن افزوده شد. پس از آنالیز های مولکولی و تایید تراریختی, مطالعات ایمنولوژیک با روشهای لکه گذاری و الیزا بر روی لاینهای تراریخت برای بررسی بیانAYT1-cMyc1 انجام شد. علاوه بر آن فعالیت استیل ترانس فرازی تراژن مذکور بر روی DON با تکنیک کروماتوگرافی لایه نازک مشاهده شد. گیاهان تراریخت تحمل نسبی به غلظت 10ppm مایکوتوکسین در ارزیابی های درون شیشه ای نشان دادند.

ازدیاد درون شیشه ای واریته city of leads رز (Rosa hybrid Cv. City of leads) از طریق کشت جوانه جانبی
یک روش تکثیر درون شیشهای کارآمد به منظور تکثیر گونه rosa hybrid واریته city of leads از طریق تکثیر جوانه جانبی بررسی گردید. در ابتدا ریزنمونهها به منظور استقرار بر روی دو محیط کشت MS و van der salm در ترکیب با غلظتهای مختلف هورمونهای BA و NAA کشت گردیدند. بیشترین درصد استقرار ریزنمونهها (100 درصد) و تعداد نوساقهها (4/3 نوساقه در هر ریزنمونه) در محیط van der salm حاوی 2 میلیگرم در لیتر BA همراه با 05/0 میلیگرم در لیتر NAA بدست آمد. در مرحله بعدی به منظور بهبود سرعت تکثیر، نوساقهها بر روی محیط کشت van der salm حاوی غلظتهای 0، 5/0، 25/1، 5/1 و 75/1 میلیگرم در لیتر BA به تنهایی و یا در ترکیب با 0، 03/0، 06/0 و 09/0 میلیگرم در لیتر NAA کشت گردیدند. بیشترین تعداد نوساقهها (6/9 نوساقه در هر ریزنمونه) بر روی محیط حاوی 75/1 میلیگرم در لیترBA همراه با 03/0 میلیگرم در لیتر NAA تولید شد. رشد طولی نوساقهها بر روی محیط van der salm حاوی 0، 10/0، 05/0 و 01/0 میلیگرم در لیتر BA به تنهایی و یا همراه با ، 03/0، 06/0 و 09/0 میلیگرم در لیتر NAA بررسی شد. بهترین تیمار جهت رشد طولی نوساقهها تیمار حاوی 01/0 میلیگرم در لیتر BA در ترکیب با 09/0 میلیگرم در لیتر NAA بود. نوساقهها به منظور ریشهزایی به محیطهای ریشهزایی حاوی هورمونهای IBA و NAA انتقال یافتند. بیشترین درصد ریشهزایی (0/93 درصد) در تیمار هورمونی حاوی 05/0 میلیگرم در لیتر NAA بدست آمد،
