ایجاد لاین های دابلهاپلویید کلزا (Brassica napus L) از طریق روش جنین زایی میکروسپور و ارزیابی قدرت ترکیبپذیری عمومی صفات مورفولوژیک و عملکردی لاین ها
در این تحقیق از هیبریدهای F1 تجاری هایولا اقدام به تولید لاینهای دابلهاپلویید کلزا (اینبرد) به روش جنینزایی میکروسپور گردید. به منظور بررسی ترکیبپذیری عمومی لاینهای دابلهاپلویید آزمون تاپکراسی با 28 لاین دابلهاپلویید و والد تاپکراس هایولا 420 انجام گرفت. هیبریدهای حاصله از تلاقی لاین دابلهاپلویید-تستر در سال زراعی 1394 کاشته شدند. صفات ارتفاع گیاه، تعداد خورجین در شاخه اصلی و فرعی، تعداد خورجین در بوته، تعداد دانه در خورجین، طول خورجین، تعداد شاخه فرعی، طول ساقه اصلی، وزن هزار دانه، عملکرد تک بوته، تعداد روز تا شروع گلدهی، تعداد روز تا خاتمه گلدهی، تعداد روز تا رسیدن فیزیولوژیک و عملکرد روغن برای ارزیابی قدرت ترکیبپذیری عمومی لاین-ها مورد تجزیه و تحلیل قرار گرفت. نتایج حاصل از تجزیه واریانس حاکی از تفاوت معنیدار بین نتاج تاپ کراس برای تمام صفات مورد بررسی در سطح احتمال یک درصد بود. بر اساس تجزیه و تحلیل مقایسه میانگینها توسط آزمون دانکن در سطح احتمال 1%، نتاج تاپ کراس شش لاین دابلهاپلویید DH1، DH8، DH10، DH11، DH13 و DH21 دارای بیشترین اختلاف معنیدار نسبت به نتاج تاپکراس سایر لاینهای دابلهاپلویید از نظر صفات مورد بررسی بودند. بیشترین میزان عملکرد بذر و عملکرد روغن مربوط به نتاج تاپکراس حاصل از تلاقی دابل هاپلویید 21 × هایولا 420 بود. بیشترین مقادیر ترکیبپذیری عمومی مثبت برای صفات عملکرد بوته، تعداد خورجین در بوته، تعداد دانه در خورجین و وزن هزار دانه مربوط به لاینهای دابلهاپلویید DH1، DH10 و DH21 بود، لذا میتوان از این سه لاین دابلهاپلویید به عنوان لاینهای والدی برتر در برنامه های اصلاحی استفاده نمود.

شناسایی ژن های مقاومت به زنگ های ساقه و قهوه ای در برخی از لاین های امیدبخش گندم با استفاده از نشانگرهای مولکولی
زنگ های ساقه و قهوه ای از خسارت زاترین بیماری های گندم در سراسر جهان می باشند. استفاده از مقاومت ژنتیکی و تولید ارقام مقاوم مطمئن ترین روش کنترل زنگها بهشمار میرود. شناسایی منابع مقاومت به زنگ و تعیین ژنهای مقاومت موجود در آنها یکی از ملزومات رسیدن به ارقام مقاوم و ایجاد مقاومت پایدار میباشد. برای تعداد قابل توجهی از ژنهای مقاومت به زنگ ساقه و قهوهای، نشانگرهای مولکولی مرتبط شناخته شده است. در این تحقیق برخی از لاینهای امیدبخش گندم در دست معرفی چهار اقلیم (گرم و مرطوب شمال، گرم و خشک جنوب، معتدل و سرد) کشور جهت تشخیص حضور ژنها و یا جایگاههای ژنی مقاومت گیاهچهای شامل Sr39/Lr35، Sr31/Yr9/Lr26/Pm8/Pm17، Sr24/Lr24 و Sr22 با استفاده از نشانگرهای مولکولی پیوسته با آنها مورد بررسی قرار گرفتند. با توجه به نتایج بهدست آمده از این تحقیق مشخص شد که جایگاه ژنی Sr39/Lr35 تنها در لاین SEP59 و جایگاه ژنی Sr31/Yr9/Lr26/Pm8/Pm17 در لاینهای S-84-14 و SEP49 وجود دارد. سایر جایگاههای ژنی در هیچ کدام از لاینهای مورد بررسی شناسایی نشدند. نتایج به دست آمده از این تحقیق نشان داد که فراوانی حضور این ژنها در لاینهای امیدبخش گندم پایین بوده و بنابراین بایستی با استفاده از لاینهای حاوی ژنهای مقاومت در برنامههای اصلاح گندم، فراوانی حضور این ژنها در ارقام اصلاح شده افزایش یابد.

شناسایی و تحلیل فیلوژنتیکی و بیانی چپرونهای هیستونی کلاس NAP در ذرت (Zea mays)
در یوکاریوتها DNA ژنومی در ترکیب با پروتئینهای هیستونی کروماتین را ایجاد میکند. چپرونهای هیستونی از طریق تغییر دسترسی به DNA بر میزان رونویسی ژنها تأثیر میگذارند. بر خلاف مخمر و جانوران، در مورد چپرونهای هیستونی گیاهی اطلاعات کمی وجود دارد. در این رابطه، خانواده nucleosome assembly protein (NAP) در تمام یوکاریوتها حفاظت شده بوده و جزء جدایی ناپذیر در پایداری، حفظ و پویایی کرماتین یوکاریوتی میباشد. این پروتئین ها در انتقال هیستونها به هسته، تشکیل نوکلئوزوم و القاء سیالیت کروماتین نقش داشته و لذا رونویسی بسیاری از ژنها را تحت تأثیر قرار میدهد. در این مطالعه با استفاده از روشهای بیوانفورماتیکی، 6 ژن شبه NAP (ZmNPL1 تا ZmNAPL6) در ذرت شناسایی شد. آنالیز فیلوژنتیکی نشان داد که این ژنهای NAPL همانند ژنهای NAPL آرابیدوپسیس و برنج به دو زیر گروه تقسیم شده و رابطه تکاملی نزدیکتری با ژنهای NAPL برنج داشتند. این ژنها دارای 3 تا 11 اینترون بوده و بر روی 5 کروموزوم از 10 کروموزوم ذرت قرار گرفتهاند. آنالیز بیانی بر پایه ریزآرایه نشان دهنده تنظیم دقیق رونویسی ژنهای ZmNAPL در طول نمو ذرت میباشد. این امر حاکی از نقش مهم این ژنها در برنامه ریزی مرتبط با فرآیندهای نموی ذرت بود. این مطالعه اولین گزارش در مورد شناسایی و بررسی روابط تکاملی، ساختاری و بیانی ژنهای NAPL ذرت بوده و نتایج بدست آمده از آن اطلاعات پایه برای تحقیقات آتی در مورد کارکرد ژن-های NAPL ذرت را مهیا میسازد.

بررسی تنوع ژنتیکی و ارتباط برای صفات مرفوفنولوژیک و مقاومت به بیماری سفیدک پودری در ژرمپلاسم گندم با استفاده از نشانگرهای ISSR، IRAP و iPBS
به منظور بررسی تنوع ژنتیکی و تجزیه ارتباط برای صفات مورفوفنولوژیک و مقاومت به بیماری سفیدک پودری در ژرمپلاسم گندم با استفاده از نشانگرهای ISSR، IPBS و IRAP ، 115 ژنوتیپ گندم در سال زراعی 94-1393 در مزرعه تحقیقاتی دانشگاه گنبد کاووس در قالب طرح بلوکهای کامل تصادفی با سه تکرار انجام شد. ژنوتیپها از تنوع بالایی برخوردار بوده و بین ژنوتیپها تفاوت معنیداری از لحاظ کلیه صفات مورد ارزیابی بجز طول دانه وجود داشت. بهمنظور ارزیابی تنوع مولکولی در 60 ژنوتیپ گندم، تکثیر از 8 آغازگر ISSR، IPBS و IRAP استفاده شد. از 61 باند تشکیل شده 47 باند چند شکل بودند. تعداد باندهای چند شکل از 2 تا 14 به ازای هر آغازگر متفاوت بود. بیشترین و کمترین درصد چندشکلی مربوط به آغازگر نوع ISSR و iPBS به ترتیب با 100 و 50 درصد بود. همچنین در این بررسی واکنش 60 ژنوتیپ و لاین گندم در برابر قارچ عامل بیماری در شرایط درون شیشهای مورد ارزیابی قرار گرفتند. تجزیه خوشهای لاینها را در سه گروه حساس، مقاوم و حدواسط قرار داد. بر اساس نتایج مقایسه میانگین به روش LSD، لاینهای 127 و 801 مقاومترین لاینها بودند در حالی که لاینهای 390، 515 و 833 بیشترین حساسیت را نسبت به بیماری نشان دادند. تجزیه ارتباط بین نشانگرها و صفات نشان داد که 56 آلل با صفات در ارتباط هستند و آلل PR50-6 بیشترین ارتباط را با صفات مورد ارزیابی داشت. با استفاده از نتایج این پژوهش ژنوتیپهای برتر میتوانند برای مراحل بعدی بهنژادی گزینش شوند.

کنترل ژنتیکی گلدهی در گیاه آرابیدوپسیس (Arabidopsis thaliana)
انتقال از رشد رویشی به فاز زایشی از تحولات مهم در زندگی گیاهان میباشد. این پدیده در گیاهان عالی تحت تاثیر بسیاری از عوامل ژنتیکی و فیزیولوژیکی میباشد. شناسایی این عوامل یکی از اهداف مهم در اصلاح بسیاری از گیاهان میباشد. در دهههای اخیر گیاه آرابیدوپسیس به عنوان یک گیاه مدل در بسیاری از مطالعات مربوط به عمل گلدهی به کار رفته است و بسیاری از مسیرهای مربوط به کنترل گلدهی در این گیاه مشخص شده است. انتقال به مرحله گلدهی توسط ژنهای ایجادکننده گل شامل FT،TSF، SOC1 و AGL24 تنظیم میشود که این ژنها شناسایی ژنهای مریستم گل را از طریق مسیرهای تناوب نوری، بهارهسازی، خودانگیزی و جیبرلین القاء مینمایند. تناوب نوری یکی از عمدهترین شرایط محیطی مؤثر در انتقال به گلدهی محسوب میشود که تحت تأثیر گیرندههای نوری فیتوکروم و کریپتوکروم و دو ژن CO و FT میباشد. ژن FLC که عمدتاً مسئول نیاز بهارهسازی در آرابیدوپسیس محسوب میشود مستقیماً به عنوان مانع تنظیم کنندههای گلدهی FT و SOC1 بوده و از انتقال به گلدهی جلوگیری میکند. ژنهای مسیر خودانگیزی عمدتاً مستقل از شرایط محیطیاند و باعث ممانعت از بیان FLC توسط فرآیند کنترل مبنی بر RNA یا تغییر کروماتین میشوند. در نهایت جیبرلین در زمانی که مسیر تناوب نوری غیر فعال است، به عنوان تسریع کننده گلدهی عمل میکند. در این مقاله مروری مکانیسم کنترل گلدهی در گیاه آرابیدوپسیس بررسی و اهمیت آن در اصلاح نباتات تشریح شده است.

بازسازی شبکههای ژنی و برهم کنش پروتئینی دخیل در پاسخ به تنش خشکی در برنج
برنج یکی از ارزشمندترین محصولات کشاورزی است. یکی از تأثیرگذارترین عوامل محدود کننده این محصول استراتژیک تنش خشکی میباشد. نظر به چند ژنی بودن تحمل به خشکی، هدف پژوهش حاضر بازسازی شبکه ژنی درگیر و شناسایی ژنهای کلیدی مربوطه در برنج با استفاده از تجزیه و تحلیل دادههای ریزآرایه بوده است. به همین منظور با استفاده از نرمافزار تحت وب Genevestigator تمامی ژنهای دارای تغییر بیان بالاتر و مساوی 5/2 و پایینتر و مساوی 5/2- در حالت تنش خشکی نسبت به شرایط نرمال در بین تمامی آزمایشات ریزآرایه انجام شده در برنج شناسایی شدند. در مجموع 101 ژن با تغییر بیان ≥5/2 و ≤5/2- شناسایی شد و شبکه تنظیم ژنی و برهمکنش پروتئینی برای آنها رسم گردید. ژنهای قطب (ژنهای دارای بیشترین برهمکنش) با استفاده از 9 الگوریتم محاسباتی Cyto-Hubba در نرمافزار Cytoscape شناسایی شدند. این امر منجر به شناسایی 14 ژن غیر تکراری شد که به عنوان مؤثرترین ژنها در پاسخ به تنش خشکی در نظر گرفته شدند و شبکه همبیانی آنها رسم گردید. بر اساس بررسی هستیشناسی ژنهای دارای بیان افتراقی، ژنهای هم بیان با آنها و ژنهای قطب، تنظیم رونویسی از گروههای اصلی بود که نشان دهنده اهمیت عوامل رونویسی در مکانیسم تحمل به خشکی میباشد. در میان عوامل رونویسی میتوان به عوامل پاسخ دهنده به تنش خشکی همچون خانوادههای ژنی AP2، bZIP، WRKY، MYB و عوامل متصل شونده به عناصر پاسخ دهنده به ABA، اشاره نمود. امید است نتایج بهدست آمده در راستای یافتن راهکارهایی هدفمند جهت افزایش تحمل به خشکی مفید واقع گردد.

ریزازدیادی Rosa canina با استفاده از مریستم جانبی
شیوه کشت درون شیشهای روش مناسبی برای تکثیر رقمهای تجاری گل سرخ با یکنواختی بسیار بالا و عاری از بیماری میباشد، امروزه به عنوان راهگشای مطلوبی در جهت اصلاح از طریق دستورزی ژنتیکی بهشمار میرود. در این مطالعه قابلیت ساقهزایی و ریشهزایی نسترن در شرایط درون شیشهای بررسی گردید. از جوانههای جانبی به عنوان ریز نمونه کشت استفاده شد. آزمایش ساقهزایی با استفاده از فاکتوریل بر پایه طرح کاملا تصادفی انجام شد. محیط کشت مورد استفاده در مرحله ساقه زایی غلظتهای مختلف تنظیم کنندههای کینتین (5/0،۰ میلیگرم در لیتر) و بنزوئیک اسید (5/0-۱-5/1-۲ میلی گرم در لیتر) بود. تجزیه واریانس برای صفات تعداد شاخساره و طول گیاهچه در سطح ۱% اختلاف معنیداری را نشان دادند. بیشترین تعداد شاخساره با غلظتهای تنظیم کننده رشد (1 میلیگرم در لیتر) بنزوئیک اسید همراه با ( 5/0 میلیگرم در لیتر) کینتین مشاهده شد. بیشترین طول گیاهچه با استفاده از تنظیمکننده-های رشد ( 5/0 میلیگرم در لیتر) BA و همراه با (5/0 میلیگرم در لیتر) Kin بهدست آمد. صفات تعداد ریشه، نسبت تعداد ریشه/طول ریشه و طول ریشه تفاوت معنیداری را در پاسخ به تیمارهای مختلف ریشه زایی نشان دادند. علاوه بر این، بهترین تیمار ریشهزایی در غلظت میلی گرم در لیتر 2/1 ایزوبنزوئیک اسید بهدست آمد. تمامی گیاهچهها سازگار شده و به گلخانه تجاری رز منتقل گردید.
