Thereby, identification of the dominant AMF species in the saline soils and their application as biofertilizer is very useful for increasing crop productivity in such conditions. For this purpose, sampling was performed from root and rhizosphere of wheat, barley and weeds in Yazd, East Azerbaijan, Qom and Markazi provinces. The morphological properties of spores of the isolated AMFs were studied. Then, samples were screened using a two steps nested PCR methodology. At the first step, AMF-specific primers, including LSU-Glom1 and SSU-Glom1 were used, followed by Alu1 restriction of PCR products, and then at the second step, the restricted PCR products were amplified by fungal universal primers (ITS4 and ITS5) for amplification of ITS-rDNA region. The PCR products were cloned, and restricted by Taq 1. The results of morphological charectreristics and analysis of the achived sequences and blasting showed that two AMF genus, including Glomus (more than 90%) and Acaulospora (10%) were domininat. The species G. mosseae (50%), G. intraradices, G. sinosum, G. constrictum, G. etunicatum, G. versiforme, G. fulvom, and Glomus sp were identified using molecular strategy. The maximum species diversity was observed in the fields of Yazd Province and rhizosphere of wheat. Totaly, results of the present study showed that the species G. mosseae has the highest dominancy and adaptivity in saline conditions, so after performing further experiments, it can be used as a source of biofertilizer in such regions.}, keywords = {Arbuscular mycorrhizal fungi (AMF),Salinity tolerance,molecular identification,Internal Transcribed Spacer (ITS),Nested PCR}, title_fa = {جدا سازی و شنا سایی قارچ‎های میکوریز آربوسکولار غالب در ریزوسفر گندم٬ جو و علف های هرز برخی مناطق زراعی شور ایران}, abstract_fa = {گیاهانی که ریشه آنها توسط قارچ‎های میکوریز آربوسکولار کلونیزه می‌شوند٬ بطور معمول نسبت به تنش‎های مختلف از قبیل شوری تحمل بالاتری دارند، بنابراین جداسازی و شناسایی این قارچ‌ها می‎تواند جهت تولید کودهای بیولوژیک برای مناطق مذکور بسیار مفید باشد. بدین منظور پس از نمونه برداری ریشه و اسپور از ریزوسفر گیاهان مورد مطالعه (گندم، جو و برخی از علف‎های هرز) از خاکهای شور استان‎های یزد، آذربایجان شرقی، قم و مرکزی، کل ژنوم گیاه و ریزوسفر با روش PCR آشیانه ای دو مرحله ای برای حضور قارچ‌های میکوریز مورد بررسی قرار گرفت. در مرحله اول PCR از آغازگرهای اختصاصی قارچ‌های میکوریزی (LSU-Glom1 and SSU-Glom1) و پس از هضم آنزیمی با Alu1، در مرحله دوم PCR از آغازگرهای عمومی قارچها (ITS5 و ITS4) استفاده شد. محصولات مرحله دوم PCR هر نمونه، همسانه سازی و کلنی‌های مثبت با آنزیم برشی Taq1 هضم شدند. با مقایسه الگوهای RFLP نمونه‌های هضم شده و توالی¬یابی نماینده‌هایی از هر الگوی برشی، وجود جنسهای Glomus و Acaulospora در ریزوسفر نمونه‌ها مشخص شد. جنس Glomus غالبترین جنس (بیش از 90 درصد) و گونه‌های Glomus mosseae(50 %)، G. intraradices ، G. versiforme، G. sinuosum ٬ G. fulvum، G. constrictum وGlomus sp در جنس مذکور مشاهده شدند. بیشترین تنوع گونه ای در خاک‌های یزد و در گیاه گندم مشاهده شد. نتایج نشان داد که گونه G. mosseae دارای بالاترین سازگاری به شرایط شور در مناطق مختلف کشور می‌باشد و لذا میتواند پس از انجام آزمایشات تکمیلی به عنوان کود بیولوژیک در این مناطق استفاده شود.}, keywords_fa = {میکوریز آربوسکولار,شناسایی مولکولی,تنش شوری,Internal Transcribed Spacer (ITS),Nested PCR}, url = {https://cropbiotech.journals.pnu.ac.ir/article_117.html}, eprint = {https://cropbiotech.journals.pnu.ac.ir/article_117_b465146f0a493942017bf728ae76022c.pdf} }
