During spring and summer of the year 2011, a total of 280 symptomatic and 463 random leaf samples, and 1186 tuber samples were collected from potato fields located in Khorasan-e-razavi and Markazi provinces, Iran and tested for PVA infection using DAS-ELISA method. Based on the results obtained, PVA infection was recorded in 39 and 11 % of symptomatic leaf samples, 23 and 6 % of random leaf samples and 17 and 13 % tuber samples in Markazi and Khorasan-e-Razavi provinces, respectively. Morever, biological properties of the selected PVA isolates were studied using host range studies on 10 plant species. Four PVA isolates were selected and their coat protein (CP) gene was amplified by specific primers using RT-PCR and their nucleotide sequences were determined (KF152950-3). The CP sequences of four Iranian and 37 Non-Iranian (from other parts of the world) PVA isolates were compared and a phylogenetic tree was reconstructed using Neighbor Joining-NJ method with 1000 bootstrap value. The PVA isolates in the phylogenic tree were clustered in two main groups (I and II) where group I had two subgroups (IA and IB). Three Iranian PVA isolates (Ar2, Kh4 and F2 all from Markazi province) were grouped in IA subgroup and showed highest similarity with two isolates from Germany and Netherlands (Y11427 and X91968) and the other Iranian PVA isolate (Gh4 from Khorasan-e-Razavi P.) was grouped in IB subgroup showing the highest similarity with an isolate from China (DQ309063). Comparison of the deduced amino acid (aa) sequences of CP among the four Iranian and the other PVA isolates revealed substitution of 2 to 7 aa mainly in N terminal of CP. This is the first phylogenetic study on Iranian PVA isolates and also the first report of PVA infection in Markazi province. This study showed variation among PVA isolates in Iran. These data could to be considered in potato breeding programs in order to achieve PVA resistant cultivars. }, keywords = {Potato virus A,detection,RT-PCR}, title_fa = {تعیین فراوانی، خصوصیات بیولوژیکی و جایگاه تبارزایی جدایه های ویروس A سیب زمینی بر اساس توالی ژن پروتئین پوششی در دو استان خراسان رضوی و مرکزی}, abstract_fa = {و ویروس A سیب‌زمینی (PVA) از جمله ویروس­های مهم خسارت‌زا در سیب‌زمینی می­باشد. در بهار وتابستان 1389، تعددا 280 و463 نمونه برگی به‌ترتیب علائم‌دار و تصادفی و نیز تعداد 1336 غده از مزارع سیب‌زمینی استان­های خراسان رضوی و مرکزی جمع­آوری وکل نمونه­ها با استفاده از آزمون الایزا از نظر آلودگی به ویروس PVA مورد بررسی قرار گرفتند. نتایج حاصل بیانگر گسترش آلودگی به این ویروس در 39 و 10 درصد نمونه­های علائم­دار، 24 و 5 درصد نمونه­های تصادفی و 17 و 13 درصد غده‌های جمع­آوری‌شده به‌ترتیب از دو استان مرکزی و خراسان رضوی بود. همچنین دامنه میزبانی جدایه­های PVA به‌دست آمده در شرایط گلخانه روی 10 جنس و گونه گیاه محک مورد بررسی قرار گرفت. با استفاده از روش RT-PCR ناحیه ژن پروتئین پوششی (CP) چهار جدایه PVA، تکثیر و توالی آن تعیین شد (رس‌شمار). توالی نوکلئوتیدی ژن CP چهار جدایه ایرانی PVA با توالی مربوطه در 37 جدایه خارجی این ویروس مورد مطالعه و مقایسه قرار گرفته و درخت تبارزایی رسم گردید. در این درخت، 41 جدایه مورد بررسی بر اساس ژن CP به دو گروه اصلی I (شامل دو زیرگروه IA و IB) و II تقسیم شدند. سه جدایه ایرانی Ar2، Kh4 و F2 از استان مرکزی در زیرگروه IA دارای بیشترین شباهت با جدایه­هایی از آلمان و هلند (به‌ترتیب Y11427 و X91968) و جدایه Gh4 در زیر گروه IB دارای بیشترین شباهت با جدایه­ای از چین (DQ309063) بود. این اولین مطالعه در زمینه روابط تبارزایی جدایه­های ایرانی PVA و نیز اولین گزارش از وقوع آلودگی به این ویروس در مزارع سیب­زمینی استان مرکزی می­باشد. نتایج حاصله نشان‌دهنده حضور واریانت­ها و جدایه­های متنوع این ویروس در کشور بوده و این اطلاعات در تهیه و اصلاح ارقام مقاوم سیب­زمینی در برابر این ویروس حائز اهمیت است. }, keywords_fa = {ویروس ای سیب¬زمینی,ردیابی,تبارزایی,واکنش زنجیره ای پلیمراز}, url = {https://cropbiotech.journals.pnu.ac.ir/article_396.html}, eprint = {https://cropbiotech.journals.pnu.ac.ir/article_396_91114d6de58f194d8545d50e932137a8.pdf} }
