In this study the alterations of SOD, GPX1 and MDHR expression level in two genotypes of rice, Pokkali (as tolerant) and IR64 (as sensitive) were monitored under different concentrations of iron levels [(0) (nonstress)], 100, 250, 400 and 500 mg/lit-1Fe-EDTA). The treatments were done when the plants were at 4-leaf stage and lasted for two weeks. Results showed that the expression levels of genes in Pokkali were higher than IR64. The expression level of SOD in IR64, increased at iron concentration increased, while it decreased at higher Fe-level. The expression level of GPX1 was increased in IR64, but decreased in Pokkali. The expression level of MDHR in IR64 was decreased at early stage of Fe-treatment, but then increased. Inversely, in Pokkali MDHR expression reduced constantly under Fe stress. Overall, the relative over expression of genes in Pokkali and presence of different expression levels of them between different concentrations of Fe in tolerant and sensitive genotypes indicate that the gene could remarkably effect on the tolerant level of pokkali by reducing ROS production under Fe-toxicity.}, keywords = {Hydroponic,IR64,Oryza sativa,Pokkali}, title_fa = {اثر سمیت آهن بر میزان بیان نسبی ژن های SOD، MDHR و GPX1 در ژنوتیپ های حساس و متحمل برنج}, abstract_fa = {افزایش تولید رادیکال‌های فعال اکسیژن در سطح سلولی در شرایط سمیت ناشی از آهن یکی از مهم‌ترین عوامل محدودکننده تولید برنج در زمین‌های کشاورزی است. در این پژوهش، تغییرات میزان بیان نسبی ژن‌های سوپراکسید‌دیسموتاز (SOD)، گلوتاتیون پراکسیداز (GPX1) و مونودهیدروآسکوربات ردوکتاز (MDHR) در دو ژنوتیپ Pokkali (متحمل) و IR64 (حساس) برنج در سطوح 0 (شاهد)، 100، 250، 400 و 500 میلی گرم در لیتر آهن (Fe-EDTA) در شرایط هیدروپونیک به مدت 2 هفته در مرحله چهار‌برگی بررسی شد. نتایج نشان داد بیان نسبی همه ژن‌های مورد بررسی در همه سطوح تنش در ژنوتیپ Pokkali بیش-تر از IR64 بود. با افزایش سطح تنش میزان بیان ژن SOD در IR64 افزایش و سپس کاهش یافت. همچنین با افزایش تنش میزان بیان ژن GPX1 در ژنوتیپ IR64 روندی صعودی و در ژنوتیپ Pokkali روندی نزولی داشت. بیان ژن MDHR در ژنوتیپ IR64 در ابتدا روند نزولی و سپس صعودی داشت. در حالی که بیان این ژن در ژنوتیپ Pokkali دارای روند کاهشی بود. به نظر می‌رسد که بیش بیانی نسبی ژن-های مورد بررسی در ژنوتیپ Pokkali نسبت به IR64 و همچنین تفاوت در روند تغییرات بیان ژن در سطوح مختلف تنش آهن می‌تواند از طریق کاهش میزان رادیکال‌های فعال اکسیژن اثر چشمگیری بر میزان مقاوت گیاه به سمیت ناشی از آهن داشته باشد.}, keywords_fa = {هیدروپونیک,IR64,Oryza sativa,Pokkali}, url = {https://cropbiotech.journals.pnu.ac.ir/article_5205.html}, eprint = {https://cropbiotech.journals.pnu.ac.ir/article_5205_9124a96d497019fc11a36e68ed744379.pdf} }
