NIa protein as a key protein in WSMV, plays important roles in viral replication and proteolytic digestion of viral polyprotein. Considering the critical role of NIa protein in viral infection, isolation, sequencing and determination of secondary and tertiary structure of this protein were the objectives of the present study. Also, the amino acids present in the NIa protein active site, which can be used as the targets for design and developement of new antiviral agents, were investigated. In this study, the gene encoding NIa protein was isolated from WSMV and its sequencing was done followed by cloning in pEST prokaryotic vector. The resulted nucleotide sequence was deposited in NCBI database with accession number MK335432. Investigation of physical and chemical properties showed that NIa protein is including 426 amino acids, 48.8 kD molecular weight and 8.81 PI. Prediction of secondary structure of NIa protein revealed that 53.29% of its structure composed of irregular loops, which was justified by the structural dirsorder of this protein. The results of the active site investigation of NIa protein based on homologous sequences alignment and three-dimensional structure of the protein showed a highly conserved site which was included of histidine, aspartic acid and cysteine amino acids.}, keywords = {Viral infection,NIa protein,Active site,Wheat,Cloning}, title_fa = {شناسایی، همسانه‌سازی و آنالیز بیوانفورماتیکی ژن کدکننده پروتئین NIa ویروس موزاییک نواری گندم (WSMV)}, abstract_fa = {ویروس موزاییک نواری گندم از مهم‌ترین آلودگی‌های گندم محسوب می‌شود که شیوع آن در ایران نیز رو به افزایش است. پروتئین NIa یکی از انواع پروتئین‌های حیاتی موجود در این ویروس است که دارای وظایف مهمی در همانندسازی، تکثیر و هضم پروتئولیتیک پلی پروتئین ویروسی می‌باشد. با توجه به نقش حیاتی پروتئین NIa در آلودگی ویروسی، هدف از مطالعه حاضر جداسازی، تعیین توالی و بررسی دقیق ساختار دوم و سوم این پروتئین بود. همچنین، آمینواسیدهای موجود در جایگاه فعال پروتئین NIa که می‌توانند به‌عنوان هدفی جهت طراحی و توسعه عوامل ضد ویروسی جدید استفاده شوند، مورد بررسی قرار گرفتند. در این مطالعه، توالی ژنی کد کننده پروتئین NIa از ویروس موزاییک نواری گندم (WSMV) ایزوله میمه، جداسازی و پس از همسانه‌سازی در ناقل پروکاریوتی pEST، تعیین توالی شد. توالی نوکلئوتیدی حاصل در پایگاه اطلاعاتی NCBI با شماره دسترسی MK335432 ثبت گردید. بررسی ویژگی‌های فیزیکی و شیمیایی پروتئین NIa نشان داد که این پروتئین دارای 426 آمینواسید، وزن مولکولی 8/48 kD و نقطه ایزوالکتریک برابر با 81/8 می‌باشد. بررسی ساختار دوم پروتئین NIa نشان داد که 29/53% از ساختار آن از لوپ‌های نامنظم تشکیل شده است، که این امر توجیهی برای وجود بی‌نظمی در ساختار این پروتئین بود. نتایج حاصل از بررسی جایگاه فعال پروتئین NIa براساس هم‌ردیفی توالی‌های همولوگ و همچنین ساختار سه‌بعدی پروتئین نشان داد که این جایگاه بسیار حفاظت شده و شامل سه آمینواسید هیستیدین، آسپارتیک اسید و سیستئین بود.}, keywords_fa = {آلودگی ویروسی,پروتئین NIa,جایگاه فعال,گندم,همسانه‌سازی}, url = {https://cropbiotech.journals.pnu.ac.ir/article_6295.html}, eprint = {https://cropbiotech.journals.pnu.ac.ir/article_6295_b41e024ec93802a2a887632d71f0823f.pdf} }
