We aimed herein to evaluate effective factors influencing the transformation efficiency and the reduction of chimeric transgenic shoots in sour lime. Epicotyl and internode explants were genetically transformed with different Agrobacterium tumefaciens strains e.g., LBA4404, GV3850, and GV3101, harboring the vectors pBI121 and pCAMBIA3301 containing β-glucuronidase (GUS) as a reporter gene. The effect of the following factors was evaluated: Agrobacterium concentration (OD600=0.3, 0.5 and 1), during inoculation (5 seconds, 10 minutes and 30 minutes), co-culture (2 and 3 days), and the selection regime (phosphinothricin at 1, 3, 5 and 10 mg/l and kanamycin at 25, 50, 75 and 100 mg/l). In following, transformation efficiency and the chimeric transgenic shoots rate were respectively confirmed by PCR and GUS assays. The results showed that Agrobacterium strain LBA4404, at the OD600 of 0.5, with 5 seconds (for epicotyl) and 10 minutes (for internode) inoculation at two-day co-culture period, were identified the most suitable treatments for both explants. The transformation frequencies ranged from 0.93% for internode on DKW medium containing 1.0 mg/l of phosphinothricin to 14.29% for epicotyl on DKW medium containing 50 mg/l of kanamycin. Inclusion of the high-level of selective treatments, improved the transformation rate through decreasing frequency escape and chimeric transgenic shoots. These findings provide novel insights into the appropriate procedure to constitute non-chimeric lime transgenic shoots.}, keywords = {GUS Reporter gene,Agrobacterium,selectable marker,chimeric tissue}, title_fa = {ارزیابی عوامل مؤثر در افزایش بازده انتقال ژن به Citrus aurantifolia L. به‌واسطه اگروباکتریوم و کاهش شیمری‌شدن شاخساره‌های تراریخته}, abstract_fa = {انتقال ژن به لیموترش (Citrus aurantifolia L.)، به‌عنوان یکی از گیاهان چوبی سرسخت، چالش‌برانگیز است. تأکید این مطالعه بر عوامل مهمی است که در افزایش موفقیت انتقال ژن و کاستن شیمری شدن شاخساره‌های تراریخته، مؤثر هستند. ریزنمونه‌های اپی‌کوتیل و میانگره، با سه سویه اگروباگتریوم (LBA4404، GV3101 و GV3850)، حامل پلاسمیدهای pBI121 و pCAMBIA3301 دارای ژن گزارشگر بتا گالاکتوزیداز (GUS)، جهت بررسی عوامل دخیل در انتقال ژن، استفاده شدند. فاکتورهای اصلی همچون OD600 اگروباکتریوم (3/0، 5/0 و 1)، زمان تلقیح ریزنمونه (5 ثانیه، 10 دقیقه و 30 دقیقه)، مدت هم‌کشتی (2 و 3 روزه) و انتخاب نوع و میزان عامل انتخابگرهای فسفینوترایسین (1، 3، 5 و 10 میلی‌گرم در لیتر) و کانامایسین (25، 50، 75 و 100 میلی‌گرم در لیتر) ارزیابی شدند. بدین منظور، بازدهی تراریختی با استفاده از واکنش زنجیره‌ای پلیمراز و میزان حالت شیمری شاخساره‌های تراریخته به‌کمک آزمون هیستوشیمیایی GUS، تأیید گردیدند. نتایج نشان داد که اگروباکتریوم سویه LBA4404، با OD600 5/0 و زمان تلقیح پنج ثانیه برای اپی‌کوتیل و 10 دقیقه برای میانگره با هم‌کشتی دو روزه، مناسب‌ترین تیمارها برای هر دو ریزنمونه بودند. فراوانی انتقال ژن بین 93/0‌% در ریزنمونه میانگره روی محیط DKW حاوی 1 میلی‌گرم در لیتر فسفینوترایسین و 29/14 درصد، در ریزنمونه اپی‌کوتیل روی محیط DKW حاوی 50 میلی‌گرم در لیتر کانامایسین حاصل شد. همچنین مشخص گردید، به‌کار بردن سطوح بالای عامل انتخابگر، بازدهی تراریزش را با کاهش پدیده شیمری و جلوگیری از فرار شاخساره‌های غیرتراریخته، بهبود می‌بخشد. یافته‌های این مطالعه، راهکارهای مؤثری را در باززایی شاخساره‌های تراریخته غیرشیمری در لیموترش، ارائه می‌دهد.}, keywords_fa = {ژن گزارشگر GUS,اگروباکتریوم,نشانگر انتخابگر,بافت شیمر}, url = {https://cropbiotech.journals.pnu.ac.ir/article_7705.html}, eprint = {https://cropbiotech.journals.pnu.ac.ir/article_7705_48ac76638de19882182fff6d52f90793.pdf} }
