SPL, the plant-specific transcription factors, are regulated by miRNAs and play important roles in several processes including tissue development, response to biotic and abiotic stress and induction of several other transcription factors and membrane proteins. In this study we selected miRNAs that regulate SPL transcription factors expression in rice. Later, the differential expression of these miRNAs are evaluated using qRT-PCR and Stem-loop primers. Results of shoot differential expression under drought stress showed that miR529 was down-regulated but conversely, miR535 was up-regulated. However, significant differential expression of miR156 was not observed in our study. Likewise, root differential expression under drought condition showed up-regulation of miR535, but miR529 and miR156 did not show any significant differential expression. Although all of these miRNAs are involved in regulating the expression of the same genes, but their diverse differential expressions highlight the complexity of gene-regulatory networks in various environmental conditions. Based on results of this study, it can be suggested that compared to miR535, miR156 and miR529 play more important roles in regulating the development and flowering process via controlling SPL transcription factors whereas, miR535, miR529 and relatively lesser miR156 are responsible for SPL transcription factor regulation under stress.}, keywords = {rice,Drought stress,miRNA,qRT-PCR}, title_fa = {بررسی تغییر بیان microRNAهای تنظیم کننده فاکتورهای رونویسی SPL برنج در پاسخ به تنش خشکی}, abstract_fa = {MicroRNA (miRNA)ها در بسیاری از فرایندهای مرتبط با تکثیر، رشد و نمو و پاسخ گیاه به تنش¬های زنده و غیرزنده نقش دارند. فاکتورهای رونویسی SPL که توسط miRNAها کنترل می¬شوند، اختصاصی گیاهان بوده و در بسیاری از فرایندهای مرتبط با نمو بافت، پاسخ به تنش¬های زنده و غیرزنده و تحریک و فعال¬سازی سایر فاکتورهای رونویسی و پروتئین¬های غشایی نقش دارند. در این تحقیق miRNAهای کنترل¬کننده تنظیم بیان فاکتورهای رونویسی SPL در برنج مشخص شدند. سپس تغییر بیان این miRNAها با استفاده از روش qRT-PCR و آغازگرهای Stem-Loop مورد بررسی قرارگرفت. نتایج نشان داد که در اندام هوایی در شرایط خشکی نسبت به نرمال، بیان miR529 کاهش و بیان miR535 افزایش یافته است؛ اما تغیر بیان معنی¬داری برای miR156 مشاهده نشد. در ریشه نیز miR535 افزایش بیان نشان داد؛ اما، miR529 و miR156 تغییر بیان معنی¬داری را در ریشه نشان ندادند. با وجود اینکه هر سه این miRNAها در کنترل ژن¬های یکسانی نقش دارند، واکنش متفاوت آنها در مواجهه با تنش خشکی نشان دهنده گستردگی شبکه¬های تنظیمی گیاه در برابر تغییر شرایط محیطی است. با توجه به نتایج می¬توان این گونه بیان کرد که miR156 و miR529 نسبت به miR535 نقش مهم¬تری را در تنظیم فرایندهای نموی و گلدهی با کنترل فاکتورهای رونویسی SPL دارند؛ در صورتی که miR535 و miR529 و به نسبت کمتر miR156 مسئول تنظیم بیان فاکتورهای رونویسیSPL در برابر تنش می¬باشند.}, keywords_fa = {برنج,تنش خشکی,miRNA,qRT-PCR}, url = {https://cropbiotech.journals.pnu.ac.ir/article_828.html}, eprint = {https://cropbiotech.journals.pnu.ac.ir/article_828_bf9fe2ac19698b525cad79f29bbd2334.pdf} }
