However, its production is extremely compromised in agricultural regions affected by water deficiency during part of the growing season and mostly in the later stages of growth. Therefore, it is promising to identify the native drought-tolerant germplasms and molecular mechanisms used to enhance drought stress resistance. The aim of this study was to investigate the expression level of eight selected genes related to drought tolerance NCED, ABF, HKT, PAL, bHLH, ABC transporter and lipoxygenase in two native germplasms of Iranian winter wheat, one is sensitive and the other is drought tolerant. For this purpose, drought treatment was applied on native germplasms in a completely randomized design with three replications and two levels of treatment in the greenhouse. Selected gene fragments were amplified, gene expression was measured by Reverse Northern Blot and quantified using total lip software. Analysis of variance of the mean relative expression of each gene compared to the internal control gene showed that drought stress had a significant effect on the expression of all genes except bHLH gene. Biplot based on the first and second components made it possible to isolate genotypes in dehydration stress based on the expression of the seven genes evaluated. This method can be used in screening and identifying tolerant genotypes in landrace population of wheat.}, keywords = {Drought stress,Gene expression,landrace population of wheat,Reverse Northern Blot}, title_fa = {بیان افتراقی ژن ‌های درگیر در مسیر تحمل به تنش کم آبی در راستای غربالگری ژنوتیپ های متحمل در گندم نان}, abstract_fa = {گندم به‌عنوان مهم‌ترین غله در بسیاری از مناطق جهان است و غذای اصلی اکثر مردم را تشکیل می‌دهد. با این وجود، اغلب مناطق تولید گندم در جهان در بخشی از فصل رشد و بیشتر در مراحل انتهایی رشد با کمبود آب مواجه هستند. لذا شناسایی ارقام متحمل و نیز مطالعه‌ی مکانیسم‌های افزایش‌دهنده مقاومت به تنش خشکی از راهکارهای مناسب جهت مقابله با عوارض تنش خشکی خواهد بود. تحقیق حاضر با هدف بررسی سطح بیان هشت ژن منتخب مرتبط با تحمل به خشکی P450 ,NCED ،ABF ، HKT ، PAL ، ABC transporter, bHLH و لیپواکسیژناز در دو ژرم پلاسم بومی‌گندم زمستانه ایرانی حساس و متحمل به خشکی انجام گرفت. سنجش بیان این ژن‌ها بر روی نمونه‌های برگی جمع‌آوری‌شده در قالب طرح کاملاً تصادفی با سه تکرار و دو سطح تیماری (نرمال و تنش خشکی) از گیاهان کشت شده در گلخانه، صورت گرفت. برای این منظور پس از تکثیر cDNA با ژن‌های منتخب، بیان آن‌ها با استفاده از روش نورترن‌بلات معکوس نسبت به یک ژن کنترل داخلی (TEF-1α) سنجیده و با استفاده از نرم‌افزار توتال لب کمی‌گردید. مطالعه بیان نسبی هر ژن نسبت به ژن کنترل داخلی نشان داد که اعمال تنش خشکی بر میزان بیان تمامی ژن‌ها به جزء ژن bHLH اثر معنی‌داری داشته‌است. نمودار دوطرفه بر پایه مولفه‌های اول و دوم، جداسازی ژنوتیپ‌ها را در تنش کم‌آبی بر اساس مقدار بیان هفت ژن مورد ارزیابی امکان‌پذیر کرد. این روش می‌تواند در غربالگری و شناسایی ژنوتیپ‌های متحمل در توده‌های بومی‌گندم کاربرد مؤثری داشته‌باشد.}, keywords_fa = {بیان ژن,تنش خشکی,توده‌های بومی‌گندم,نورترن بلات معکوس}, url = {https://cropbiotech.journals.pnu.ac.ir/article_8930.html}, eprint = {https://cropbiotech.journals.pnu.ac.ir/article_8930_415c015cee741d0b9a126999a6edc308.pdf} }
