تأثیر نانواکسید تیتانیوم و متیل جاسمونات بر میزان محتویات فنلی و فلاونوئیدی کل در کشت سوسپانسیون سلولی گیاه کبر (Capparis spinosa)

چکیده

این پژوهش به منظور بررسی تولید کالوس، کشت سوسپانسیون سلولی و تأثیر القا کننده بر میزان محتویات فنل و فلاونوئیدی کل در گیاه کبر (Capparis spinosa) انجام شد. بدین منظور، ریزنمونه‌های مختلف (برگ و ساقه) از گیاهان بالغ جمع‌آوری و پس از ضدعفونی، در شرایط استریل کشت داده شدند. اثر تیمارهای مختلف هورمونی شامل ترکیباتNAA و BAP بر القای کالوس مورد ارزیابی قرار گرفت. ریزنمونه‌ها در محیط کشت MS حاوی غلظت‌های مختلف این تنظیم‌کننده‌های رشد کشت شدند. بر اساس نتایج بیشترین درصد کالوس‌زایی (100 درصد) در محیط کشت حاوی 1 میلی‌گرم بر لیتر2,4-D و 5/0 میلی‌گرم بر لیترBAP حاصل شد. در مرحله بعدی اثرنانو ذرات دی اکسید تیتانیوم /و متیل جاسمونات به عنوان الیستور بر محتویات فنلی و فلاونوئیدی کل در کشت سوسپانسیون سلولی این گیاه مورد بررسی قرار گرفت. نتایج حاصل از تجزیه واریانس داده های مربوط به میزان فنل و فلاونوئید کل حاصل از کشت سوسپانسیون سلولی گیاه کبر نشان داد اثر‌متقابل نانو ذرات دی اکسید تیتانیوم و متیل جاسمونات در سطح احتمال 99 درصد معنی‌دار شده است. بیشترین میزان فنل کل (80/60 میلی‌گرم در گرم ماده خشک) و فلاونوئید کل (36/49 میلی‌گرم در گرم ماده خشک) از کاربرد نانو اکسید تیتانیوم به میزان 25 میکروگرم در لیتر و متیل جاسمونات به میزان 100 میکرومول در محیط کشت سوسپانسیون حاصل شد. این یافته‌ها می‌توانند در جهت بهینه‌سازی تولید ترکیبات زیست‌فعال گیاه کبر در شرایط درون شیشه و تحت کنترل مورد استفاده قرار گیرند.

چکیده [English]

This study was conducted to investigate callus production, cell suspension culture and the effect of induction on total phenolic content in Capparis spinosa. For this purpose, different explants (leaves and stems) were collected from adult plants and after disinfection, they were cultured under sterile conditions. The effect of different hormonal treatments including NAA and BAP compounds on callus induction was evaluated. The explants were cultured in MS medium containing different concentrations of these growth regulators. The results showed that the highest callus induction percentage (100%) was obtained in medium containing 1 mg/L 2,4-D and 0.5 mg/L BAP. The results of the variance analysis of data related to the amount of total phenol and flavonoids obtained from the cell suspension culture of the caper plant showed that the interaction effect of titanium dioxide nanoparticles and methyl jasmonate was significant at the 99% probability level. The highest level of total phenols (60.80 mg/g DW) and total flavonoids (49.36 mg/g DW) were related to the use of nano-titanium oxide 25 μg/L and methyl jasmonate 100 μmol as elicitor in the suspension culture medium. These findings can be used to optimize the production of bioactive compounds from C. spinosa under controlled in vitro conditions.
