Soybean (Glycine max L.) is one of the most important agricultural products in the world, and its breeding using biotechnology methods is one of the effective strategies to overcoming the problems of this plant. Achieving an efficient regeneration protocol is an important step in gene transfer programs into plants. The aim of this investigation was to optimize some effective factors in organogenesis of soybean. For this purpose, the effect of some factors on organogenesis of soybean such as the age of cotyledon explants (6, 8, 10 and 12 days), the type and concentration of gelling agents such as agar (4, 6, 8 and 10 g/l) and gelrite (2, 2.5 and 3 g/l) and finally different levels of the ethylene inhibitor silver nitrate (0, 2.5, 5 and 10 mg/l), were investigated independently. All experiments were performed in a completely randomized design with three replications. The results showed that there was a negative correlation between explant age and organogenesis, so that the highest number of shoots per explant (3.5) was obtained from 6-day-old cotyledon explants. And with the increase of explant age from 6 days to 12 days, the number of shoots per explant (0.8) decreased significantly. Also, agar and its concentration of 6 g/L were introduced as the most suitable gelling agent for direct organogenesis of soybean. The presence of silver nitrate had a significant positive effect on organogenesis traits, such that the highest number of shoot per explant (6.8) was observed at a concentration of 5 mg/L of silver nitrate.
