Two key genes involved in the biosynthetic pathway of this compound are 1-deoxy-D-xylulose 5-phosphate reductase(DXR) and gamma-terpinene synthase(GTS). In this study, changes in the expression of the DXR and GTS genes were investigated in two Iranian black cumin ecotypes with the highest(H) and lowest(K) levels of thymoquinone, under the influence of silver nitrate nanoparticles(AgNPs) treatment. The plants were treated with AgNPs at a concentration of 20mg/L during the immature seed stage, and sampling was conducted at 12, 24, and 48 hours after treatment. The relative expression levels of the genes were evaluated using the RT-qPCR method. The results showed, at a 5% significance level, that the application of AgNPs for 48 hours led to an increase in the expression of the GTS gene in both ecotypes. Furthermore, the results indicated that the expression level of the DXR gene was affected differently by AgNPs in the two ecotypes. In ecotype H, the expression of the DXR gene significantly decreased compared to the control, with the lowest expression observed at 48hours. In ecotype K, the expression of the DXR gene initially increased significantly at 12hours and then exhibited a decreasing trend, with the lowest expression recorded 24hours after treatment. Overall, the findings of this research suggest that AgNPs treatment can alter the expression patterns of the GTS and DXR genes in the thymoquinone biosynthetic pathway in black cumin plants; however, the manner and extent of these changes are significantly influenced by the plant genotype and the timing of treatment application.
