بررسی تنوع ژنتیکی لاین های جهش یافته آفتابگردان (Helianthus annuus L.) و شناسایی نشانگرهای AFLP پیوسته با صفات گیاهچه ای تحت شرایط تنش شوری

چکیده

تنش شوری از جمله تنش های غیر زنده تهدید کننده گیاهان است. مطالعه تنوع ژنتیکی گیاهان برای پیشبرد برنامه‌های به نژادی گیاهی در راستای ایجاد ارقام متحمل به تنش مهم می‌باشد. در این تحقیق، ابتدا واکنش گیاهچه ای 53 لاین جهش یافته آفتابگردان به همراه والد مادری در سطوح مختلف شوری (صفر، 2، 4، 6 و 8 دسی‌زیمنس‌بر‌متر) بررسی شد. همچنین، در آزمایش مولکولی تنوع ژنتیکی لاین ها با استفاده از نشانگرهای AFLP مطالعه گردید. نتایج نشان داد که از نظر صفات گیاهچه ای اختلاف معنی دار بین لاین های مورد مطالعه در هر یک از سطوح شوری وجود دارد. با استفاده از 17 ترکیب آغازگری AFLP، تعداد 200 مکان ژنی در جمعیت مورد مطالعه تکثیر یافت. با تجزیه ساختار جمعیت مورد مطالعه، لاین های جهش یافته مورد مطالعه در 2 زیر گروه قرار گرفتند. نتایج مکان یابی ارتباطی نشان داد که تعداد 43 نشانگر ارتباط معنی دار با صفات گیاهچه ای دارند. ضریب تبیین فنوتیپی نشانگرهای شناسایی شده بین 13/0 تا 42/0 متغییر بود.

چکیده [English]

Salinity stress is one of the abiotic stresses which threaten plant species. Achieving any progress in breeding for tolerance to stresses needs existence of genetic diversity. In this research, reaction of 53 sunflower mutant lines seedlings accompanied by their parental line was studied in several salinity levels including 0, 2, 4, 6, 8 ds/m. Also, in molecular experiment, genetic diversity of mutant lines was inspected using AFLP markers. Results revealed significant differences among the studied lines in each of salinity level based on seedling characteristics. Two hundreds loci were amplified using 17 combinations of AFLP primers in the studied lines. Analysis of population structure classified studied mutant lines in two subgroups. Considering association mapping, 43 AFLP markers had significant relation with seedling characteristics. Phenotypic variance of the identified markers varied between 0.13 and 0.42.
