چکیده

سفیدک داخلی (Pseudoperonospora cubensis (P. cubensis یکی از مهمترین بیماری‌های گیاه خیار است که موجب کاهش تولید آن در جهان می‌شود. با وجود در دسترس بودن بعضی از قارچ‌کش‌ها برای کاهش خسارت این بیماری، تمایل به استفاده از محصولات طبیعی جدید و قابل اطمینان افزایش یافته است. در این پژوهش اثر دو ماده اسید سالیسیلیک (SA) و اسید آزلائیک (AZA) با غلظت ی‍ک میلی‌مولار بر برخی خصوصیات بیوشیمیایی و بیان برخی ژن‌های دفاعی در گیاه خیار به‌صورت آزمایش اسپلیت - فاکتوریل در قالب طرح کاملاً تصادفی در سه تکرار مورد مطالعه قرار گرفت. بررسی بیان ژن نشان داد با وجود این‌که بعد از مایه‌زنی گیاهان با قارچ، بیان هر دو ژن کیتیناز بتا-1 و 3- گلوکاناز به‌طور معنی‌داری افزایش یافت، مقدار بیان ژن کیتیناز در گیاهان تیمارشده با SA و سطح بیان ژن بتا-1 و 3- گلوکاناز در گیاهان تیمارشده با AZA بیشتر بود. همچنین نتایج این بررسی نشان داد که مقدار کلروفیل a، b و فلاونوئید در گیاهان تیمارشده با AZA و SA در 6 ساعت بعد از مایه‌زنی با قارچ در مقایسه با شاهد بیشتر بود. مقدار فنل کل هم در گیاهان تیمارشده با SA و AZA نسبت به شاهد بیمار به‌ترتیب درساعات 6 و 72 بعد از تیمار افزایش یافته بود. این نتایج نشان می‌دهد که پیش تیمار گیاهان خیار با القاگر‌های SA و AZA موجب پاسخ‌های سریع‌تر و افزایش کارایی سیستم دفاعی در برابر بیماری سفیدک داخلی می‌شود.

چکیده [English]

Downy mildew is one of the most important diseases of cucumber plants reducing production in the world. Despite the availability of some fungicides to control the disease, demands for new and reliable natural products has been increased. In this study, the effects of salicylic acid (SA) and Azelaic acid (AZA) on some biochemical properties and expression of defense genes in cucumber plants were studied. Gene expression analysis indicated that expression of both β-1,3- glucanase and chitinase genes were increased after inoculation with Pseudoperonospora cubensis, but this increase was much higher for chitinase gene in SA-treated plants and for β-1,3- glucanase in AZA-treated plants. Also, chlorophyll a, b and total flavonoids contents in AZA and SA-treated plants were higher compared to control 6 hours after inoculation with pathogen. Total phenolic content was also increased in SA and AZA treated plants compared to the control 6 and 72 hours after treatment, respectively. These findings indicate that pretreatment of cucumber plants with SA and AZA prompts faster defense responses and increases the efficiency of the defense system against downy mildew.
