بررسی تنوع ژنتیکی پروانه جوانه خوار بلوط Tortrix Viridana (Lep. Tortricidae) درجنگل های بلوط زاگرس با استفاده از s rDNA28

چکیده

پروانه جوانه خوار بلوط Tortrix viridana (Lep. Tortricidae)، یکی از آفات مهم جنگل‌های زاگرس است. به منظور کنترل شیوع این آفت و جلوگیری از بروز خسارت به درختان بلوط زاگرس که از لحاظ اقتصادی دارای اهمیت ویژه‌ای هستند، بررسی تنوع ژنتیکی جمعیت‌های پروانه جوانه خوار بلوط در جنگل‌های بلوط شمال غرب ایران، انجام شد. نمونه‌های مورد بررسی T. viridana از گونه میزبان بلوط Quercus branti و از مناطق جنگلی استان‌های آذربایجان غربی، لرستان، کردستان و کرمانشاه جمع آوری شدند. به منظور بررسی تنوع ژنتیکی از ژن rDNA 28s هسته‌ای با طول bp 850 استفاده شد. نمونه‌برداری از قسمت سر و تنه حشره کامل انجام شد. استخراج DNA به روشCTAB انجام گرفت و سپس توالی‌یابی قطعه ژنی هسته‌ای28S rRNA انجام شد. توالی‌های DNA با استفاده از نرم افزار Bioedit ویرایش شدند و همردیفی با نرم افزار MegaX و درخت فیلوژنتیک به روش neighbor-joining با تکرار 1000 و با نرم افزار MegaX ترسیم شد. نتایج درخت فیلوژنتیک نشان داد که نمونه‌های مختلف پروانه جوانه خوار بلوط بر اساس فاصله جغرافیایی دارای تنوع ژنتیکی هستند. ارزیابی ساختار ژنتیکی جمعیت‌ها و برسی شاخص‌های Fst و Nm نشان داد که به دلیل جریان ژنی کم مابین جمعیت‌ها، تنوع ژنتیکی بین جمعیت‌ها بیشتر از درون جمعیت‌ها است. هم‌چنین مارکر 28s چون جزء مارکرهای حفاظت شده می‌باشد توانسته به خوبی بین جمعیت‌ها تفرق ایجاد کند و تنوع ژنتیکی را به درستی نشان دهد. نتایج این تحقیق می‌تواند در به‌کارگیری روش‌های مورد استفاده شیمیایی و بیولوژیک جهت کنترل این آفت مورد استفاده قرار گیرد.

the genetic diversity of the T. viridana on the host oak species Quercus branti was investigated using the 28s gene sequence in some Zagros forests. The samples were collected from the forest area of West Azarbaijan, Lorestan, Kurdistan and Kermanshah provinces. The Collected samples, which were in the larval stage, were grown in laboratory conditions until they turned into paup and then into complete insects, for DNA extraction. DNA extraction was done by CTAB method. 21 samples were selected to investigate genetic diversity using the 28s gene, and 18 sequences DNAs were of suitable quality for further investigations. In order to amplify 28s region, based on the sequence obtained from the NCBI site.The desired region in the collected samples was amplified using the PCR method and the resulting product was sequenced. The sequences were edited using Bioedit software and aligned with the help of MegaX software, and the phylogenetic tree was drawn by UPGMA method with 1000 sampling repetitions. The results of this study, based on the phylogenetic tree, showed that the different samples of the oak saplings have genetic diversity based on the geographical distance. Assessment of genetic structure of the populations showed that higher diversity was demonstrated in intrapopulations than interpopulations geographically. 28s gene showed the presence of 5 haplotypes in these populations. The presence of low diversity between populations indicated the existence of gene flow between populations as well as genetic compatibility in the studied populations.
